Trifluridine: A Fluorinated Pyrimidine Nucleoside with Anti-Herpesvirus and Antitumor Activities

Trifluridine (trifluorothymidine, TFT), a fluorinated pyrimidine nucleoside, is an anti-herpesvirus agent and an antitumor antimetabolite agent. Thymidylate kinase (TK) is a key enzyme in the synthesis of thymidine-5'-monophosphate [T0845, di-Na salt], an essential precursor for DNA biosynthesis. Trifluridine is incorporated into (virus or tumor) DNA as its triphosphate via phosphorylation by thymidylate kinase (TK), ultimately suppressing DNA replication and inhibiting cell proliferation.1-4) Trifluridine has activity against herpes simplex virus types 1 and 2 (HSV-1 and HSV-2), and vaccinia virus.1-3) Recently, a combined antitumor agent TAS-102 (trifluridine with tipiracil, a thymidine phosphorylase inhibitor) has been used in clinical. Inclusion of tipiracil increases trifluridine exposure by inhibiting its metabolism by thymidine phosphorylase. 4-6) (The product is for research purpose only.)
