Bortezomib: A Reversible Inhibitor of the 26S Proteasome in Mammalian Cells
Bortezomib (PS-341) is a reversible inhibitor of the 26S proteasome in mammalian cells. The proteasome is an abundant multi-enzyme complex that provides the main pathway for degradation of intracellular proteins in eukaryotic cells. The 26S proteasome is a large protein complex that degrades ubiquitinated proteins. The ubiquitin-proteasome pathway plays an essential role in regulating the intracellular concentration of specific proteins. The orderly degradation of cellular proteins is critical for normal cell cycling and function, and inhibition of the ubiquitin-proteasome pathway with bortezomib results in cell-cycle arrest and apoptosis.
