Application
Amphotericin B: A Polyene Macrolide Antifungal Agent
Amphotericin B is a polyene macrolide antifungal agent, and acts by binding to the sterol component, ergosterol, of the cell membrane of susceptible fungi. The mechanisms of action has been reported that it forms transmembrane channels leading to alterations in cell membrane permeability through which monovalent ions (Na+, K+, H+, and Cl-) leak out of the cell resulting in cell death. Amphotericin B shows a high order of in vitro activity against many species of fungi which are all inhibited by concentrations of it ranging from 0.03 to 1.0 μg/mL in vitro. Amphotericin B is also effective against leishmaniasis, a parasitic disease caused by the Leishmania parasite. Recently, in order to improve the tolerability of amphotericin B and reduce toxicity, several formulations, such as liposomal formulations, have been developed.
